To our knowledge, this study is the first report on yeast tolerance mechanisms against imidazoles, such as miconazole, and their mode of action using a genome-wide screening approach in yeast. However, miconazole and fluconazole are characterized by different activity features: fluconazole does not seem to induce ROS to the same extent as miconazole in susceptible fungi 2, 3 ; . In this study, we screened the haploid set of Saccharomyces cerevisiae deletion mutants in non-essential genes for both hypersensitivity and resistance to miconazole, by determining the minimal inhibitory miconazole concentration MIC ; for all individual yeast knock-out mutants using two-fold dilution series of miconazole in liquid YPD medium. In this way, no miconazoleresistant yeast mutants could be identified. However, we could identify 29 S. cerevisiae genes.

It was demonstrated that polymorphisms of TNF-, IL-10, IL-6, and IFN- modulate the synthesis of these cytokines. The aim of our study was to analyze relationship between genotypes, of above mentioned polymorphisms, and kidney function up to 36 months after successful cadaveric kidney transplantation. Methods: Genotyping was performed in 240 subsequent cadaveric kidney recipients transplanted in one center from 1998 to 2002. Genomic DNA was obtained from peripheral leukocytes. Identification of cytokine genotypes was based on PCR-SSP method for TNF- at position -308 A G ; , IL-10 at positions -1082 A G, -819 T C, -592 A C ; , IL-6 at position -174 G C ; and IFN- at position + 874 T A ; . Genotypes were grouped according to the strengths of expression of cytokines. Graft function was analyzed after 12, 24 and 36 months. In 18 out of 240 patients primary graft non-function PGN ; was observed. During the follow up period 33 patients underwent graftectomy due to chronic allograft nephropathy CAN ; and 12 patients died with functioning kidney allograft. Patients with PGN and those who died were excluded from survival analyses. Results: A faster, significant decline of kidney graft function was observed in patients with genotypes: TT IFN- + 874 T A ; , GG and GC IL-6 174 G C ; , and GG TNF 308 A G ; . Only in case of Il-6 polymorphism its' influence on graft survival was confirmed. According to the KaplanMeier method the graft survival was in patients with GG and GC genotypes determinating higher IL-6 production ; significantly shorter worse F-Cox test p 0.02 ; than in patients with CC genotype lower IL-6 synthesis ; . We did not observe any significant influence of IL-10 polymorphisms -1082 A G, -819 T C, -592 A C ; on graft function or survival. Conclusions: IL-6 promotor G174C genotype CC is associated with better kidney graft survival, and genotype TT of INF- intron 1 or genotype GG of TNF- promoter with faster decline of graft function than any other genotypes of this cytokine, for instance, .

As illustrated in Figure 1, the 24-reductase catalyses many reactions, but their relative sequence has not been established so far because of lack of definitive studies on the substrate specificity of the enzyme. We based the selection of the 24 25 ; -ene sterol intermediates on work reported from other laboratories [5, 8, 12, 21, that suggests that C-24 reduction occurs either before or after, but not during, C30, C-31 and C-32 demethylation of lanosterol. We therefore prepared only essential representative sterol intermediates : lanosterol, zymosterol, 5-cholesta-7, 24-dien-3-ol and desmosterol. Also, in a previous work16 82% of the Candida strains, isolated from patients with various diseases of the respiratory tract, were identified as a C. albicans. Strains susceptibility tests showed that 5-fluorocytosine was the best antimycotic in vitro, in a similar manner as was observed in our study. We suggest that they may be interpreted as three C. albicans strains subpopulations: a susceptible one MIC 1.56 mg ml ; , an intermediate susceptible MIC 3.125-12.5 mg ml ; and a resistant one 45% of the strains; MIC 25-50 mg ml ; . These results are similar to that reported by others, who found a threemodal miconazole MIC distribution for Candida sp. strains. 15 MICs values for resistant, intermediate susceptible, and susceptible subpopulations were 32 mg ml, 8.1-32 mg ml, and 8 mg ml, respectively. 15 Therapeutic options often are limited in patients with azole resistance oropharingeal and esophageal candidiasis, particularly when there is cross-resistance to other antifungal triazoles. Intravenous amphotericin B may be the only alternative antifungal compound available to treat esophageal candidiasis and advanced oropharingeal candidiasis, although, amphotericin B resistance has been described.5, 17.

Two teams of federal and university scientists announced that they had resurrected the 1918 influenza virus and found that it was actually a bird flu that jumped directly to humans. Democrats on Capitol Hill are complaining that the delay has put Americans in jeopardy. Tamiflu, introduced in 1999, has recently become the drug of choice of the medical establishment worried about pandemic flu because it is one of the only medicines claimed to reduce the duration and severity of the potentially deadly disease if taken within 48 hours of infection. Tamiflu is `supposed' to speed recovery from the flu. When started during the first 2 days of the illness, it is said to hasten improvement by at least "a day." PRINCIPLES OF ASSESSMENT Presenting symptoms of AD may be vague and are often easily missed especially when impairment is mild and accommodated for by a spouse or other family member, behavioral problems are minimal or other medical conditions demand more attention. Studies show that in primary care a diagnosis of AD is made relatively late in the course of illness although symptoms have been evident for several years. Abstract Objecti e: To evaluate the efficacy of Neo-Penotran pessaries Zmetronidazole 500 mgq miconazole nitrate 100 mg. in candidal, bacterial and trichomonal vaginitis and in mixed vaginal infections. Method: Ninety-seven patients with clinical diagnosis of vaginitis entered this open, non-comparative study. Each patient inserted one pessary twice daily for 7 days. Gynecological and microbiological assessments were carried out before, and 8 10r21 23 days after the start of treatment. Results: Vaginitis symptoms were resolved in 91% of the 74 patients evaluated, and improved in a further 7%. Microbiological cure rates were 97.3% for trichomonal, 86.6% for bacterial and 81% for candidal vaginitis. Recurrence rates were 2.7, 3.8 and 16.1%, respectively. Overall microbiological cure rate for mixed infections was 86%, with 93% for trichomonal q bacterial, and 73% for bacterial q candidal vaginitis. In two out of three cases with trichomonal q bacterial q candidal infection, the microorganisms were eradicated completely. Conclusion: Neo-Penotran provides immediate and effective treatment for vaginitis, irrespective of single or multiple infection, even when the diagnosis may be uncertain. 2001 International Federation of Gynecology and Obstetrics.
